RP-HPLC-RI-PDA Method Development and Estimation of Sucrose, Fructose, and L-Ascorbic Acid in Pear Juice

Abstract


To simultaneously quantify and analytically differentiate sucrose, fructose, and L-ascorbic acid in commercial samples of Pyrus communis L. fruit juice, the research paper presents an analytical methodology based on High-Performance Liquid Chromatography (HPLC) with Refractive Index Detectors (RI) and Photodiode Array Detectors (PDA). Juice from pears included sucrose, fructose, and L-ascorbic acid.  The study outlines an HPLC RI-PDA approach that is quick, easy, sensitive, picky, and reliable and may be used to measure sucrose and fructose in pear juice. The suggested analytical approach was verified, and the findings showed good linearity, accuracy, and precision. The accurate UV-spectroscopy method for assessing sucrose, fructose, and ascorbic acid was shown to have good linearity in the drug method validation. The devised method uses the analytical wavelengths of 270nm, 340nm, and 290nm to measure sucrose, fructose, and L-ascorbic acid. The developed HPLC-RI-PDA method was successfully applied to quantify sucrose, fructose, and L-ascorbic acid in commercial pear juice samples. This analytical method would contribute valuable knowledge on methodological approaches and analyzing critical nutritional components in fruit juices.
